Introduction  

We conducted a yearly polluted-reference sampling to assess the effects of petroleum pollution on life cycle characteristics of the meiobenthic nematode Odontophora villoti. Samples were taken every 15 days between 26 November 2004 and 25 November 2005 from two beaches of Bizerte bay (Tunisia), Rimel and Tunisian Refining Industries Company (TRIC). Enteric viruses monitoring in surface waters requires the concentration of viruses before detection assays. The aim of this study was to evaluate different methods in terms of recovery efficiencies of bacteriophage PP7 of Pseudomonas aeruginosa, measured by real-time PCR, using it as a viral control process in water analysis. Different nucleic acid extraction methods (silica–guanidinium thiocyanate, a commercial kit (Qiagen Viral RNA Kit) and phenol–chloroform with alcohol precipitation) exhibited very low recovery efficiencies (0.08–4.18 %), being the most efficient the commercial kit used for subsequent experiments. To evaluate the efficiency of three concentration methods, PBS (as model for clean water) and water samples from rivers were seeded to reach high (HC, 106 pfu ml?1) and low concentrations (LC, 104 pfu ml?1) of PP7. Tangential ultrafiltration proved to be more efficient (50.36?±?12.91, 17.21?±?9.22 and 12.58?±?2.35 % for HC in PBS and two river samples, respectively) than adsorption–elution with negatively charged membranes (1.00?±?1.34, 2.79?±?2.62 and 0.05?±?0.08 % for HC in PBS and two river samples, respectively) and polyethylene glycol precipitation (15.95?±?7.43, 4.01?±?1.12 and 3.91?±?0.54 %, for HC in PBS and two river samples, respectively), being 3.2–50.4 times more efficient than the others for PBS and 2.7–252 times for river samples. Efficiencies also depended on the initial virus concentration and aqueous matrixes composition. In consequence, the incorporation of an internal standard like PP7 along the process is useful as a control of the water concentration procedure, the nucleic acid extraction, the presence of inhibitors and the variability of the recovery among replicas, and for the calculation of the sample limit of detection. Thus, the use of a process control, as presented here, is crucial for the accurate quantification of viral contamination.  相似文献

Intensive land development as a result of the rapidly growing tourism industry in the “Riviera Maya” region of the Yucatan Peninsula, Mexico may result in contamination of groundwater resources that eventually discharge into Caribbean coastal ecosystems. We deployed two types of passive sampling devices into groundwater flowing through cave systems below two communities to evaluate concentrations of contaminants and to indicate the possible sources. Pharmaceuticals and personal care products accumulated in the samplers could only have originated from domestic sewage. PAHs indicated contamination by runoff from highways and other impermeable surfaces and chlorophenoxy herbicides accumulated in samplers deployed near a golf course indicated that pesticide applications to turf are a source of contamination. Prevention and mitigation measures are needed to ensure that expanding development does not impact the marine environment and human health, thus damaging the tourism-based economy of the region.  相似文献

Extracts of 19 of 24 species of sponge collected from Queensland. Australia, inhibited the growth of test microorganisms in in-vitro assays. A similar result has been obtained by earlier workers for extracts of species of sponge obtained from temperate waters. Some of the extracts, including extracts of Thorecta vasiforis, Arenochalina mirabilis and Acanthella kleutha, showed activity against bacterial strains that was comparable with that exhibited by penicillin G and streptomycin against these strains. Gram-positive bacteria were expecially sensitive to many extracts. Little activity was exhibited by any extract against four species of fungi tested. Some of the extracts were markedly toxic to one or more of the test organisms (a fish, a crustacean and a hydroid) used, but no clear pattern linking toxicity to these organisms with antimicrobial properties of the extracts emerged. In general, there bas a negative correlation between antimicrobial activity and surface-fouling, raising the possibility of using freedom from surface-fouling as an indicator of antimicrobial activity. Four of five encrusting species from the undersides of coral boulders showed antimicrobial activity. This study confirmed the value of using methanol-toluene extracts in field-based screening programmes, but emphasised the need to use additional extracting media such as methylene chloride in order to augment the number of antimicrobial compounds detected. There are indications that antimicrobial activity may be widespread in the order Dictyoceratida, in the order Verongida and in the order Axinellida, but sporadic in other orders of Porifera.  相似文献

The most widely used technique for abatement of acid drainage from inactive surface mines and refuse disposal areas is revegetation of a soil cover applied to the waste material. Nonetheless, acid production often persists and in some cases, limits establishment of vegetation. This paper reports on several field studies intended to determine the location of pyrite oxidation zones and migration pathways of oxidation products at inactive spoil and refuse sites.Oxygen required for pyrite oxidation is believed to he provided in the gaseous state from the atmosphere. Therefore, the oxygen concentration in unsaturated mine waste should provide an estimate of the weathering tendency in the local environment. We are currently monitoring gas composition in refuse and spoil at six sites. Barren refuse appeared to be oxygenated (>2% 02) in a shallow zone extending less than 1 metre below the surface during most of the year. Preliminary data from coal spoil showed that oxygen can be available throughout the unsaturated thickness, even at a revegetated site. Gas composition varied vertically and laterally at a single site and also appeared to show seasonal dependence.Hydrologic factors are also important in acid production and transport. Discharge monitoring alone does not adequately describe the mass transport of acid products through the spoil. For example, at one reclaimed mine the mean sulfate content in six monitoring wells ranged from 24% to 240% of the mean concentration at the discharge point. Sources of recharge and relative flow rates determine the contribution of a particular zone to overall discharge quality.These basic studies of acid production and transport indicate some shortcomings of standard reclamation practices at certain sites. This information will he used to develop alternative abatement technology designed to mitigate acid production at the source.  相似文献

Colloid-facilitated phosphorus (P) delivery from agricultural soils in different hydrological pathways was investigated using a series of laboratory and field experiments. A soil colloidal P test was developed that yields information on the propensity of different soils to release P attached to soil colloids. The relationship between turbidity of soil extracts and total phosphorus (TP) was significant (r2 = 0.996, p < 0.001) across a range of agricultural soils, and a strong positive relationship (r2 = 0.86, p < 0.001) was found between "colloidal P" (H2O-CaCl2 extracts) and turbidity. Linear regression of the proportion of fine clay (<2 microm) for each soil type evaluated against the (H2O-CaCl2) colloidal P fraction gave a weak but positive relationship (r2 = 0.38, p = 0.082). The relative contribution of different particle-size fractions in transporting P in agricultural runoff from grassland soils was evaluated using a randomized plot experiment. A significant difference (p = 0.05) in both TP and reactive phosphorus (RP) in subsurface flow was recorded for different particle-size fractions, with most TP transferred either in association with the 2-microm fraction or with the 0.001-microm or smaller fractions. Total P concentrations in runoff were higher from plots receiving P amendments compared with the zero-P plots; however, these differences were only significant for the >0.45-microm particle-size fractions (p = 0.05), and may be evidence of surface applications of organic and inorganic fertilizers being transferred through the soil either as intact organic colloids or attached to mineral particles. Our results highlight the potential for drainage water to mobilize colloids and associated P during rainfall events.  相似文献

The relationship of natural marsh-estuarine systems to the economic productivity of marine systems is not well understood, at least in any quantitative sense. An approach is developed for relating blue crab economic productivity on Florida's Gulf Coast to marsh availability in the area. Previous efforts have not always applied economic concepts appropriately in attempts at such quantification. The marginal value productivity of marsh is shown to vary with alternative levels of marsh and effort in the fishery. The interaction and subsequent interdependence is shown to be statistically significant. Data availability on marginal response to marsh changes poses a severe obstacle to further progress.  相似文献
